To get the ball rolling, my 2 favorites:

Blade Runner: Rutger Hauer soliloquy ("tears in rain")

Contact:the 18 minute journey

Darth Vader to Lando in Empire Strikes Back:

"I have altered the deal, pray I do not alter it further"

The perfect line befitting one of the great villains in all cinema.

Another excellent scene, this one from Terry Gilliam's prophetic classic Brazil:

Sam Lowry returns to his apartment to find that two civil-service technicians have trashed it, all in retaliation for his earlier refusal to fill out a form.

Two shuttles lifting off simultaneously and flying into space 200 yards apart to rescue us from "Armageddon." Definitely a goose-bump moment.

Especially for those of us who prize scientific accuracy. NASA people use that film as a trainer for spotting errors and so far the count is 168.

Hill Street Blues about 25 years ago: cop has just expounded on the benefits of modern society when the pop machine refuses to dispense his purchase. As he is beating on it another character says: "Open the pod bay doors, Hal."

Animal House. Bluto: "Christ, seven years of college down the drain." I know it's not Sci Fi, but it might as well have been.

Whole 9 Yards

Jimmy T: "Oz, it's not important how many people I've killed. What's important is how I get along with the people who are still alive"

Mission Control: Buckaroo, The White House wants to know is everything ok with the alien space craft from Planet 10 or should we just go ahead and destroy Russia?
Buckaroo Banzai: Tell him yes on one and no on two.
Mission Control: Which one was yes, go ahead and destroy Russia... or number 2?

Dunno about great, but here's one of the stupidest, from a movie called "Earthstorm". (click on photo link.) I had just taken a big gulp of Pepsi when this scene came on and I ended up squirting it all out my nose and started choking with laughter.
